Transport and metabolism of choline in synaptosomes: ionic requirement.

چکیده
100pM-choline and incubated with lOOp~-[Me-~'C]choline only one energy of activation was observed (40.0kJ/mol) over the whole range of temperatures studied (6-40°C; Table 1). Similarly in nerve-cell cultures it was observed that, after incubation with concentrations of choline analogous to the ones present in the growth medium (30m~) , only one energy of activation was found (Massarelli et al., 1979). The results should be correlated with the changes in the endocellular choline pool when the synaptosomes are incubated in the absence of choline (Massarelli & Wong, 1980) and indicate that after preincubation in the absence of choline and incubation with very low concentrations of choline (lphi) the energy of activation showed a value not exceeding 29kJ/mol, which is rather low compared with other active-transport processes. Below 23-25OC, when the synaptosomal membranes are known to change their state, the uptake and the transport of choline appear to require more energy. This does not happen when the endocellular concentrations of choline are increased after preincubation and incubation with 1 0 0 ~ ~ choline.

The 'compartmentation' of choline acetyltransferase within the synaptosome.
1. Choline acetyltransferase may be isolated in either a bound or soluble form after hypo-osmotic treatment of a crude synaptosome fraction, depending on the conditions. 2. In the bound form, the enzyme appears to be associated with the larger membrane fragments rather than with synaptic vesicles. 3. The bound form is predominant at slightly acid pH values and low ionic strength, the soluble fo...
متن کاملInhibition of acetylcholine synthesis and tyrosine nitration induced by peroxynitrite are differentially prevented by antioxidants.
Evidence of an overload of reactive oxygen species and peroxynitrite, a derivative of nitric oxide, in sporadic amyotrophic lateral sclerosis suggests that peroxynitrite could impair cholinergic functions. Because of the impossibility of obtaining synaptosomes from vertebrate neuromuscular junctions, we used cholinergic synaptosomes purified from Torpedo marmorata electroneurons to characterize...
متن کاملIonic requirements of [3H]GBR-12935 binding to the dopamine transporter in canine striatal membranes.
The action of the neurotransmitter dopamine, following its synaptic release is terminated by reuptake into the nerve terminals [I]. This process is dependent on sodium and mediated by a membrane located transporter [2]. The transporter has been implicated as a cocaine receptor related to drug selfadministration [3]. The dopamine transporter has been studied directly via the accumulation of radi...
متن کاملAn efficient one-pot synthesis of α-amino nitriles using ecofriendly Lewis-acidic ionic liquid choline chloride.2ZnCl2
α-Amino nitriles are synthesized in a one-pot, three-component coupling of aldehydes (or ketones), aniline, and trimethylsilyl cyanide using a catalytic amount of choline chloride.2ZnCl2 as a bio Lewis acidic ionic liquid. Mild Lewis acidic characteristic of this ionic liquid allows efficient synthesis of α-amino nitriles from acid sensitive aldehydes.
متن کاملEffect of Paraoxon on GABA Uptake by Rat Cerebral Cortex Synaptosomes
Background: It has been suggested that organophosphates may inhibit gamma-aminobutyric acid (GABA) metabolism in synaptosomal preparations. In the present investigation, we have assessed the interaction between paraoxon and the GABA system at synaptic level. Methods: Synaptosomes were prepared from male Wistar rats (200-250 g). Cerebral cortex was dissected and homogenized, then centrifuged at ...
